Thousands of animal and plant species have evolved to produce various toxins and alkaloids designed either to defend themselves or attack and kill their prey. Many of these compounds are deadly, while others cause some highly unpleasant conditions and symptoms, such as pain, inflammation, itch, nausea, vomiting etc. During millions of years of evolution these molecules have been “polished” to become highly potent and selective – a yet to be achieved dream of the pharmaceutical research and development. In many cases, voltage- and ligand-gated ion channels represent the molecular targets of these natural compounds highlighting multiple vital roles of ion channels. We will discuss the usefulness of natural toxins as pharmacological tools for the study of ion channels, characterization of their biological functions, as well as an important source of major medications, including first-in-class agents, for the treatment of many diseases from cardiovascular and metabolic disorders to chronic pain.
